目的:观察通痹散对体外培养的强直性脊柱炎(AS)人成纤维细胞成骨的药理作用。
方法:采用2例AS患者的髋关节囊,体外培养AS成纤维细胞,以中药血清药理学方法制备不同浓度的通痹散含药大鼠血清,分别通过MTT法、羟脯氨酸(HYP)比色法、125I标记放射免疫(RIA)法测定其对AS成纤维细胞的增殖、分泌胶原和骨钙素的影响。
结果:不同浓度的通痹散含药大鼠血清对AS成纤维细胞的增殖、分泌胶原和骨钙素均有抑制作用(P<0.05),且呈剂量依赖性。
结论:通痹散对AS成纤维细胞的成骨潜能有明显的抑制作用。 |

| Effect of Tongbi powder(通痹散)on osteogenesis of cultured fibroblasts obtained from patients with ankylosing spondylitis |

ABSTRACT
Objective:To study pharmacological actions of Tongbi powder(通痹散)on osteogenesis of cultured fibroblasts obtained from patients with ankylosing spondylitis(AS).
Methods:Fibrous tissues were obtained from capsula articularis coxae of AS patients after replacement of total hip,and fibroblasts were cultured in vitro.The drug-containing serum of the rats treated with different doses of Tongbi powder(通痹散)was prepared by using serum pharmacological method of Chinese herbs.MTT assay,HYP determination and 125I radioimmunoassay methods were adopted to study effects of Tongbi powder(通痹散)on proliferation capacity,synthesis of collagen and osteocalain(OCN)of the third generation of AS fibroblasts cultured in vitro.
Results:Three different doses of drug-containing serum greatly inhibited the proliferation capacity and the synthesis of both collagen and OCN of AS fibroblastsl(P<0.05),and the inhibition was in a dose-dependent manner.
Conclusion:Tongbi powder(通痹散)could inhibit the ostiogenic ability of AS fibroblasts. |
